It checks for fetal blood cells in the mother's system and is usually done only when excessive fetal transfusion is suspected. It determines if the mother needs more than one injection of Rhogam. The Kleihauser-Betke stain could also be used to determine whether fetal transfusion has taken place in any Rh- mother of an Rh+ baby, in order to avoid an unnecessary injection of Rhogam.
